Inhalt | Basics Genetic polymorphisms; allelic variants; haplotypes; genotypes; phenotypes; allelic frequencies; ethnic group frequencies: Odds-ratio; positive predictive factors; negative predictive factors; genome-wide association studies; expression priofiles; etc. Pharmaco- /Toxicogenetics Pharmacogenetics in clinical setting; personalized medicine; gene-based dosie selection/adjustement; gene-based patient selection; companion tests. Expression genetics Gene expression profiles in cellular systems; gene expression profiles in preclinical animal models; gene expression proflles in the clinical staging of diseases and/or disease outcome under selected therapy regimen; realization of diagnostic tests. |
Lernziele | The students understand the basic principles of pharmaco- and toxicogenomics. They also understand the concepts of pharmaco- and toxicogenetics and of expression genetics. The students understand the meaning of the correlation of genotype (or haplotype) and phenotype and corresponding tests aiming at optimizing of drug therapies. The students know a selection of important genes (and their most frequent allelic variants) which play major roles as efficacy targets for selected therapies, as targets of impaired drug disposition, and as targets for predispositions of patients for serious adverse drug effects. The students understand the meaning of expression profiles and, based on selected examples, the use of such profiles in the preclinical and clinical development of drugs as well as in the prediction of disease progression and/or therapy outcomes. The students understand the meaning of selected alleles and/or their expressed protein variants as biomarkers and their usage in as diagnostics in the approval process of new therapeutics. |
